Abstract:
The objective is to improve the methodology for conducting species inventories in the monitoring of the biodiversity of the waterbodies from Macun (SNP). It will be focused on the use of eDNA analyses (metabarcoding). Such method is particularly well adapted for alpine ponds, because of their closed water area (without upstream contamination) and also of their low species richness (linked to altitude). An eDNA session (sampling, metabarcoding) will be conducted in 2021 on five ponds, targeted to invertebrates, amphibians, fishes, mammals. A traditional sampling (with net) will also be conducted, with the objective to compare the data obtained by both methods. Furthermore, the database collected by net sampling in Macun ponds between 2002 and 2019 will also be taken into consideration. The outcome will be an appraisal of the contribution of eDNA analyses for species inventories in Macun ponds, with potentially propositions for an adaptation of the protocol used since 2002
